A fatal case of urosepsis due to Corynebacterium riegelii BJM
Aygun,Gokhan; Midilli,Kenan; Cilingir,Hatice; Yilmaz,Mesut; Kutukcu,Aysegul; Eker,Engin.
Corynebacterium species other than Corynebacterium diphtheriae rarely cause infections in human but rather reside in flora, however they have been reported to cause opportunistic infections in both immunocompromised and immunecompetent patients. Here we report for the first time a case of an elderly female patient presenting with a fatal urosepsis caused by a recently defined pathogen, Corynebacterium riegelii, identified on second day after patient hospitalization leading to a progressive worsening and death of the patient on 6th day.

A feeding association between Wilson's Storm-petrels Oceanites oceanicus (Kuhl, 1820) and Rough-toothed Dolphins Steno bredanensis (G. Cuvier in Lesson, 1828) Biota Neotropica
Olmos,Fábio; Rotenberg,Elsie; Muscat,Edelcio.
On 06 October 2012 a pod of Rough-toothed Dolphins Steno bredanensis (Delphinidae) was observed catching and tearing apart several Castin Leatherjacks Oligoplites saliens (Carangidae) near Montão de Trigo Island, northern coast of São Paulo, Brazil. The resulting floating masses of viscera attracted a feeding aggregation of 120-150 Wilson's Storm-petrels Oceanites oceanicus (Hydrobatidae), a transequatorial migrant passing through the area during October. This interspecific association has not been recorded before, although it could be expected given that S. bredanensis feeds on large fish and O. oceanicus is a known scavenger of kills made by cetaceans such as Killer Whales.

A high-fructose diet induces changes in pp185 phosphorylation in muscle and liver of rats BJMBR
Ueno,M.; Bezerra,R.M.N.; Silva,M.S.; Tavares,D.Q.; Carvalho,C.R.; Saad,M.J.A..
Insulin stimulates the tyrosine kinase activity of its receptor resulting in the tyrosine phosphorylation of pp185, which contains insulin receptor substrates IRS-1 and IRS-2. These early steps in insulin action are essential for the metabolic effects of insulin. Feeding animals a high-fructose diet results in insulin resistance. However, the exact molecular mechanism underlying this effect is unknown. In the present study, we determined the levels and phosphorylation status of the insulin receptor and pp185 (IRS-1/2) in liver and muscle of rats submitted to a high-fructose diet evaluated by immunoblotting with specific antibodies. Feeding fructose (28 days) induced a discrete insulin resistance, as demonstrated by the insulin tolerance test. Plasma...

A holoparasitic plant severely reduces the vegetative and reproductive performance of its host plant in the Caatinga, a Brazilian seasonally dry forest Acta Botanica
Cruz Neto,Oswaldo; Leal,Inara Roberta; Santos,Jean Carlos; Lopes,Ariadna Valentina.
ABSTRACT Host-parasite interactions between plants may reduce the vegetative and reproductive performance of the host plant. Although it is well established that parasitic plants may negatively affect the metabolism and the number of vegetative/reproductive structures of their hosts, the effects of this interaction on the reproductive characteristics of the host plant are poorly understood. Here we document the interaction between Cuscuta partita (Convolvulaceae) and its main host, Zornia diphylla (Fabaceae), in the Caatinga of northeastern Brazil. We measured diverse reproductive/vegetative attributes of Z. diphylla in 60 plots randomly distributed in patches that were parasitized and not parasitized by C. partita. Both vegetative and reproductive...

A laboratory cage for foster nursing newborn mice BJMBR
Marques-de-Araújo,S.; Cardoso,M.A..
We describe a cage to be used for foster nursing in order to guarantee that original mother's colostrum is not ingested by the newborn mice. A common (30.5 cm x 19.5 cm x 12.0 cm) mouse cage was fitted with a wire net tray with a mesh (1 cm x 1 cm), which divides the cage into an upper and a lower compartment. Mice born to females placed in the upper compartment pass through the mesh and fall into the lower compartment, where another lactating female with one or two of its own pups are. Of a total of 28 newborn mice of C3H/He and Swiss strains, 23 were successfully fostered. Important observations are presented to show that this is a valuable alternative for foster studies without great suffering on the part of the female.

A leucine-supplemented diet improved protein content of skeletal muscle in young tumor-bearing rats BJMBR
Gomes-Marcondes,M.C.C.; Ventrucci,G.; Toledo,M.T.; Cury,L.; Cooper,J.C..
Cancer cachexia induces host protein wastage but the mechanisms are poorly understood. Branched-chain amino acids play a regulatory role in the modulation of both protein synthesis and degradation in host tissues. Leucine, an important amino acid in skeletal muscle, is higher oxidized in tumor-bearing animals. A leucine-supplemented diet was used to analyze the effects of Walker 256 tumor growth on body composition in young weanling Wistar rats divided into two main dietary groups: normal diet (N, 18% protein) and leucine-rich diet (L, 15% protein plus 3% leucine), which were further subdivided into control (N or L) or tumor-bearing (W or LW) subgroups. After 12 days, the animals were sacrificed and their carcass analyzed. The tumor-bearing groups showed a...

A liquid phase blocking ELISA for the detection of antibodies against infectious bronchitis virus BJMBR
Cardoso,T.C.; Mouro-Sousa,R.L.; Oliveira,C.; Stringhini,G.; Augusto-Pinto,A..
A liquid phase blocking ELISA (LPB-ELISA) was developed for the detection and measurement of antibodies against infectious bronchitis virus (IBV). The purified and nonpurified virus used as antigen, the capture and detector antibodies, and the chicken hyperimmune sera were prepared and standardized for this purpose. A total of 156 sera from vaccinated and 100 from specific pathogen-free chickens with no recorded contact with the virus were tested. The respective serum titers obtained in the serum neutralization test (SNT) were compared with those obtained in the LPB-ELISA. There was a high correlation (r2 = 0.8926) between the two tests. The LPB-ELISA represents a single test suitable for the rapid detection of antibodies against bronchitis virus in...

A liver metalloendopeptidase which degrades the circulating hypotensive peptide hormones bradykinin and atrial natriuretic peptide BJMBR
Carvalho,K.M.; Nava,R.A.; França,M.S.F.; Medeiros,M.A.S.; Camarão,G.C.; Juliano,L..
A new metalloendopeptidase was purified to apparent homogeneity from a homogenate of normal human liver using successive steps of chromatography on DEAE-cellulose, hydroxyapatite and Sephacryl S-200. The purified enzyme hydrolyzed the Pro7-Phe8 bond of bradykinin and the Ser25-Tyr26 bond of atrial natriuretic peptide. No cleavage was produced in other peptide hormones such as vasopressin, oxytocin or Met- and Leu-enkephalin. This enzyme activity was inhibited by 1 mM divalent cation chelators such as EDTA, EGTA and o-phenanthroline and was insensitive to 1 µM phosphoramidon and captopril, specific inhibitors of neutral endopeptidase (EC 3.4.24.11) and angiotensin-converting enzyme (EC 3.4.15.1), respectively. With Mr 85 kDa, the enzyme exhibits optimal...

A mathematical framework for group analysis of von Willebrand factor multimeric composition following luminography BJMBR
Lopes,A.A.; Soares,R.P.S.; Maeda,N.Y..
The objective of the present study was to establish a method for quantitative analysis of von Willebrand factor (vWF) multimeric composition using a mathematical framework based on curve fitting. Plasma vWF multimers from 15 healthy subjects and 13 patients with advanced pulmonary vascular disease were analyzed by Western immunoblotting followed by luminography. Quantitative analysis of luminographs was carried out by calculating the relative densities of low, intermediate and high molecular weight fractions using laser densitometry. For each densitometric peak (representing a given fraction of vWF multimers) a mean area value was obtained using data from all group subjects (patients and normal individuals) and plotted against the distance between the peak...

A micromethod for quantitation of debrisoquine and 4-hydroxydebrisoquine in urine by liquid chromatography BJMBR
Pereira,V.A.; Auler Jr.,J.O.; Carmona,M.J.; Mateus,F.H.; Lanchote,V.L.; Breimer,D.D.; Santos,S.R.C.J..
We describe a new simple, selective and sensitive micromethod based on HPLC and fluorescence detection to measure debrisoquine (D) and 4-hydroxydebrisoquine (4-OHD) in urine for the investigation of xenobiotic metabolism by debrisoquine hydroxylase (CYP2D6). Four hundred µl of urine was required for the analysis of D and 4-OHD. Peaks were eluted at 8.3 min (4-OHD), 14.0 min (D) and 16.6 min for the internal standard, metoprolol (20 µg/ml). The 5-µm CN-reverse-phase column (Shimpack, 250 x 4.6 mm) was eluted with a mobile phase consisting of 0.25 M acetate buffer, pH 5.0, and acetonitrile (9:1, v/v) at 0.7 ml/min with detection at lexcitation = 210 nm and lemission = 290 nm. The method, validated on the basis of measurements of spiked urine, presented 3...

A moderate decrease in temperature inhibits the calcium signaling mechanism(s) of the regulatory volume decrease in chick embryo cardiomyocytes BJMBR
Souza,M.M.; Boyle,R.T..
Chick cardiomyocytes, when submitted to hyposmotic swelling, exhibit a partial regulatory volume decrease (RVD). A Ca2+ influx by stretch-activated channels signals a taurine efflux and the RVD at 37°C. We evaluated the cell's performance at room temperature. Cardiomyocytes isolated and cultured from 11-day-old chick embryos were submitted to a hyposmotic solution (180 mOsm/kg H2O) at 37°C and at room temperature (26°C). Under these conditions we measured the changes in cell volume as well as the intracellular free Ca2+ (using fura-2). During hyposmotic swelling, cells at 37°C displayed a peak relative volume of 1.61 ± 0.03 and recovery to 1.22 ± 0.04 (N = 14), while cells at 26°C presented a peak swell relative volume of 1.74 ± 0.06 and did not recover...

A molecular marker distinguishes the subspecies Melipona quadrifasciata quadrifasciata and Melipona quadrifasciata anthidioides (Hymenoptera: Apidae, Meliponinae) Genet. Mol. Biol.
Waldschmidt,Ana M.; Barros,Everaldo G. de; Campos,Lucio A.O..
The stingless bee species Melipona quadrifasciata includes two subspecies, Melipona quadrifasciata anthidioids and Melipona quadrifasciata quadrifasciata. The morphological difference between the two subspecies is the presence of three to five continuous yellow stripes on the terga on the 3rd to 6th segments in workers and males of M. q. quadrifasciata, and two to five interrupted bands in M. q. anthidioides. We identified a DNA marker which is present in M. q. quadrifasciata and absent in M. q. anthidioides. Only one among the M. q. quadrifasciata colonies did not present the marker. It was also absent in bees collected in northern Minas Gerais State (Brazil), despite their morphological resemblance to M. q. quadrifasciata. The marker can be used for...
